    Title: 養殖型脈指形軟珊瑚sinularia gibberosa 活性成分對癌細胞抗腫瘤活性及誘發細胞凋亡之分子機制研究

    Abstract: 此研究由養殖型Sinularia gibberosa軟珊瑚所分離出的六種活性物質flaccidoxide、flaccidoxide-13-acetate、11,12-Epoxy-13,14-dihydroxycembrene-C、gibberoketosterol、gibberoketosterol B 及(1E,3E,7E,11E)-(14R)-cembra-1,3,7,11-tetraen-14-ol,其生理活性評估及其對細胞之分子機制則尚未被探討。在此計畫之初步實驗中發現,flaccidoxide 及flaccidoxide-13-acetate 對於不同人類癌細胞具有抑制生長之作用;flaccidoxide 會造成肝癌細胞產生凋亡現象,並由實驗中發現其促進肝癌細胞所產生之細胞凋亡,可能經由粒線體功能喪失,引起粒線體膜電位改變,使cytochrome C釋放至細胞質中並活化caspase3/9。研究中也發現flaccidoxide會活化Unfolded Protein Response (UPR)訊息傳遞途徑,並且經由PERK/eIF2/ ATF4/CHOP 途徑,造成內質網壓力延長,使得細胞生長停滯並造成細胞凋亡。另外研究也發現mitogen-activated protein kinase (MAPK)訊息途徑也可能參與flaccidoxide誘導肝癌細胞產生細胞凋亡,此研究已經完成。另一研究進行flaccidoxide及flaccidoxide-13-acetate對膀胱癌細胞之研究,目前已經發現flaccidoxide對於膀胱癌細胞會造成其產生細胞凋亡之效果,並且可以經由降低PI3K/Akt/mTOR訊息傳遞途徑機制所產生。
