    Title: 探討預發芽糙米萃取物改善第二型糖尿病之作用機制

    Abstract: 我們先前的研究已經發現給予預發芽糙米(Pre-germinated brown rice , PGBR)可以改善因高脂飼料所導致的高血糖,但我們並不知道是何物質影響了血糖。因此本研究目的是以預發芽糙米萃取物(PGBRE)是否可以預防高脂飼料所導致小鼠的高血糖。
    將六週大的SHR大鼠隨機分組,每組8隻。我們利用SD rat先以低劑量 STZ 35mg/kg及nicotinamide 200mg/kg腹腔注射,隔日再進行一次同樣的注射,並以高脂肪飼料(60% fat, 18.6% protein and 21.4% carbohydrate, as a percentage of total kcal)混合0.5%膽固醇, 餵食4週,若飯前血糖值超過300mg/dl及胰島素無明顯改變,即為第二型糖尿病 (Hussein et al., 2011)。確定第二型糖尿病誘導成功後進行分組,一組持續餵食高脂飼料,一組食用高脂飼料同時每日口服PGBR萃取物(0.5, 5, 10 mg/kg)。比較高脂組與實驗組,結果發現高脂組有較高的血糖與胰島素濃度;且高脂組老鼠肝臟的AMP激活的蛋白激酶(AMP-activated protein kinase , AMPK)、胰島素受體(insulin receptor B, IRB)、胰島素受體基質-2 (insulin receptor substrate-2, IRS-2)、serine/threonine kinase PI3K-linked protein kinase B (Akt)、葡萄糖運輸體蛋白(2 glucose transporter-2, GLUT-2) 及葡萄糖激酶 (glucokinase, GCKR) 的表現異常。然而口服PGBRE的實驗組,其血糖濃度,胰島素濃度有明顯的改善。另外,PGBRE能增加AMPK, IR, IRS-2, Akt, GLUT-2 及GCKR 的蛋白表現。
    綜觀本實驗的結果,PGBRE可以預防STZ及nicotinamide所引發的第二型糖尿病,其可能的作用機制是經由促進胰島素的敏感與調節路徑及促進葡萄糖代謝。
